学术报告: Metabolic Engineering of microorganisms for the production of 3-hydroxypropionic acid from glycerol

内容简介:Biodiesel can be produced from plant oil, animal fats and or microalgal biomass. For every 9 kg of biodiesel production, about 1 kg of crude glycerol is generated as a byproduct. Glycerol is a good carbon source for the production of 3-hydroxypropionic acid (3-HP),a valuable platform chemical. Several genetically engineered microbes such as Escherichia coli, Klebsiella pneumoniae and Pseudomonas denitrificans have been developed for the biological production of 3-HP by introducingan aldehyde dehydrogenase (ALDH; NAD+-dependent)anda glycerol dehydratase (GDHt; vitamin B12-dependent). However, both the enzymes were seriously inactivated during the reaction due to damage in coenzyme B12 and/or accumulation of the toxic intermediate 3-hydroxypropionaldehyde (3-HPA). The loss of enzyme activity reduced 3-HP production rate, especially in the late stage of fermentation. To minimize the inactivation of the enzymes and maintain their activities at proper levels, balanced expression of the genes encoding the pathway enzymes was attempted by promoter engineering including 5’UTR modification and bicistronic design (BCD) strategy. In addition, unique dynamic promoters which are induced by 3-HP have been employed. The dynamic promoters could express the genes of the two enzymes continuously and maintained their activity until the end of bioreactor runs. This talk summarizes our long efforts for the 3-HP production and discusses the potential and limitations of our work. Also, glycerol metabolisms affecting 3-HP production in these microbes are discussed.

韩国生物技术和生物工程BBE论文最佳引用奖(2016),第三十界韩国生物技术和生物工程年会特别贡献奖(2015)等。研究方向:生物催化与生物转化、生物制氢、生物基化学品与手性化学品的生物制造、群体感应。
